  • PCR實驗室布局及通風系統特點介紹

    PCR實驗又稱基因擴增實驗,每個實驗區必須相互獨立,不能直接連接。每個獨立實驗區都設有專門的大門,并有供工作人員更換工作服和工作鞋的區域。那么PCR實驗室應該如何布局呢?下面跟著小編一起來了解下吧!

    臨床基因擴增檢驗實驗室原則上分為四個分隔開的工作區域:
    The clinical gene amplification testing laboratory is generally divided into four separated work areas:
    1、試劑貯存和準備區
    1. Reagent storage and preparation area
    2、標本制備區
    2. Specimen preparation area
    3、擴增反應混合物配制和擴增區
    3. Preparation of amplification reaction mixture and amplification zone

    如使用全自動分析儀,區域可適當合并,各工作區域必須有明確的標記,避免不同工作區域內的設備、物品混用。進入各工作區域必須嚴格按照單一方向進行,即試劑貯存和準備區→標本制備區→擴增反應混合物配制和擴增區→擴增產物分析區。
    If a fully automatic analyzer is used, the areas can be appropriately merged, and each working area must have clear markings to avoid mixing equipment and items in different working areas. Entering each work area must strictly follow a single direction, namely the reagent storage and preparation area → specimen preparation area → amplification reaction mixture preparation and amplification area → amplification product analysis area.
    PCR實驗室可以是分散形式,也可以是組合形式。完成一組PCR實驗,通常應經過試劑配制、樣品處理、核酸擴增及產物分析四個實驗過程,若實驗工藝需要,還應增加樣品粉碎過程。
    PCR laboratories can be in dispersed or combined forms. Completing a set of PCR experiments usually involves four experimental processes: reagent preparation, sample processing, nucleic acid amplification, and product analysis. If the experimental process requires it, the sample crushing process should also be added.
    核酸擴增室及產物分析室應呈微負壓,以防含核酸的氣溶膠擴散出去污染試劑與樣品,可以通過控制排風風量大于進風風量達到負壓效果。在理想情況下,PCR實驗室緩沖間內,可設置正壓,使室內空氣不流向室外,室外空氣不流向室內。 PCR實驗室進風由原有空調控制,要求將空調風口安裝到指定定點,且高度為地面鋪裝好后2600mm處。
    The nucleic acid amplification room and product analysis room should have a slight negative pressure to prevent the diffusion of aerosols containing nucleic acid from contaminating reagents and samples. The negative pressure effect can be achieved by controlling the exhaust air volume to be greater than the inlet air volume. In an ideal scenario, a positive pressure can be set in the PCR laboratory buffer room to prevent indoor air from flowing to the outside and outdoor air from flowing indoors. The inlet air of the PCR laboratory is controlled by the original central air conditioning, and it is required to install the central air conditioning air outlet at the designated location, with a height of 2600mm after the ground is paved.
    若房間進深允許,可設PCR內部專用走廊。需要指出的是在減少室內外空氣交換方面,緩沖間比專用走廊較有意義。
    If the room depth allows, a dedicated PCR internal corridor can be set up. It should be pointed out that in reducing indoor and outdoor air exchange, buffer rooms are more meaningful than dedicated corridors.
